Overview

AOD-9604 (Anti-Obesity Drug 9604) is a synthetic 16-amino acid peptide fragment derived from the C-terminal domain of human growth hormone (hGH), specifically residues 177-191 with a tyrosine substituted for phenylalanine at the N-terminal end. It was originally developed by Professor Frank Ng at Monash University, Australia in the 1990s with the objective of isolating the lipolytic domain of hGH - the region responsible for fat metabolism - while eliminating the anabolic and diabetogenic effects associated with full-length growth hormone administration.

The defining research claim for AOD-9604 is its selectivity: it is proposed to stimulate lipolysis and inhibit lipogenesis via beta-3 adrenergic receptor upregulation in adipose tissue without activating hGH receptors, without elevating IGF-1 levels, and without affecting glucose homeostasis or insulin sensitivity. In preclinical animal models, this profile was confirmed. However, this selective efficacy did not translate fully into large-scale human clinical trials, and development was terminated in 2007 following a 24-week trial of 536 subjects that failed to demonstrate sufficient weight loss for regulatory approval.

A separate and important regulatory distinction: the FDA has granted AOD-9604 "Generally Recognized As Safe" (GRAS) status when used as a food additive - a designation that evaluates safety, not efficacy, and does not imply approval for therapeutic use. AOD-9604 remains an investigational compound with no approved therapeutic indication.

Mechanism: Upregulates beta-3 adrenergic receptor expression in adipose tissue, stimulating lipolysis. Inhibits lipogenesis (conversion of non-fat food to stored fat). Does not bind hGH receptors, does not elevate IGF-1, and does not affect glucose or insulin homeostasis in preclinical models.

Development History: Multiple Phase I and Phase II human trials conducted across approximately 900 participants with a well-characterised safety profile. Storage Requirements

AOD-9604 is supplied as a lyophilised solid. The disulfide bridge between Cys(7) and Cys(14) is critical to the peptide's structural integrity and proposed biological activity. Reducing conditions or highly oxidising environments during reconstitution can compromise this structural feature. Handling should be performed with standard non-reducing buffers and the reconstituted solution should not be exposed to strong oxidising agents.

Preparation Notes

  1. Allow the sealed vial to reach room temperature before opening to minimise moisture introduction.
  2. Use bacteriostatic water (BAC water) as the reconstitution solvent. Add solvent slowly down the inside wall of the vial. Use a non-reducing buffer - do not use DTT or beta-mercaptoethanol-containing buffers as these will reduce the critical disulfide bridge.
  3. Gently swirl until fully dissolved. Do not vortex. AOD-9604 is soluble in aqueous solutions and dissolves readily at room temperature.
  4. Once reconstituted, store at 2-8°C. Use within 28 days. Do not freeze reconstituted solution - the disulfide bond may be compromised by repeated freeze-thaw cycling in solution.
  5. Discard if solution appears cloudy, discoloured, or contains particulate matter. AOD-9604 solution should be clear and colourless.
