Overview

Epithalon (also rendered as Epitalon or Epithalone) is a synthetic tetrapeptide consisting of four amino acids: alanine, glutamic acid, aspartic acid, and glycine (sequence: Ala-Glu-Asp-Gly). It was derived from Epithalamin, a crude polypeptide extract obtained from the bovine pineal gland, and developed through research led by Professor Vladimir Khavinson at the St. Petersburg Institute of Bioregulation and Gerontology beginning in the 1980s. The synthetic peptide was confirmed to be present in physiological pineal gland extract for the first time in 2017, establishing its natural origin.

Over the subsequent 25 years, Epithalon has been evaluated across a substantial body of in vitro, in vivo, and in silico studies, as well as clinical trials focused primarily on gerontological outcomes. Research has concentrated on its reported influence on telomerase activation, circadian rhythm regulation via melatonin pathways, antioxidant defence, and neuroendocrine modulation. It remains one of the most extensively studied synthetic bioregulatory tetrapeptides in the gerontology literature.

Origin: Derived from Epithalamin, a polypeptide extract of the bovine pineal gland. The synthetic sequence replicates the active tetrapeptide fragment (AEDG) identified as responsible for the parent extract's bioregulatory properties.

Research Context: Studied in cell cultures, animal models, and clinical cohorts, primarily in a gerontological context. Research spans telomere biology, circadian regulation, immune function, and antioxidant pathways. Human clinical data exists but is predominantly from Russian research institutions.

Storage Requirements

Epithalon is supplied as a lyophilised (freeze-dried) solid. As a tetrapeptide, it has relatively good stability compared to larger peptides, but correct handling remains essential to preserve compound integrity. Follow the specifications below.

Temperature: -20°C Long-term storage. For short-term use (up to 4 weeks), 2-8°C is acceptable.

Light Sensitivity: Light-Sensitive. Store in original opaque vial. Avoid direct UV or prolonged exposure to light.

Shelf Life: 24 Months. Lyophilized, sealed, stored at -20°C. Batch expiry printed on vial label.

Preparation Notes

  1. Allow the sealed vial to reach room temperature before opening to minimise moisture introduction.
  2. Use bacteriostatic water (BAC water) as the reconstitution solvent. Add solvent slowly down the inside wall of the vial - do not inject directly onto the lyophilised cake.
  3. Gently swirl the vial until fully dissolved. Do not vortex or shake vigorously. Epithalon dissolves readily in aqueous solution.
  4. Once reconstituted, store at 2-8°C. Use within 28 days. Do not freeze reconstituted solution.
  5. Discard if solution appears cloudy, discoloured, or contains particulate matter.
