Overview

GHRP-2 (Growth Hormone Releasing Peptide-2), also known by its INN pralmorelin and development code KP-102, is a synthetic hexapeptide and potent agonist of the ghrelin/growth hormone secretagogue receptor (GHS-R1a). It was developed through the pioneering work of endocrinologist Cyril Y. Bowers in the 1980s and 1990s as part of a systematic programme to identify peptide analogues capable of stimulating GH release. GHRP-2 is regarded as the most potent member of the classical synthetic GHRP family by peak GH output.

GHRP-2 operates through two complementary mechanisms at the pituitary level: it amplifies the natural GH-releasing signal via GHS-R1a activation, and simultaneously suppresses somatostatin (growth hormone-inhibiting hormone), removing the primary inhibitory brake on GH secretion. This dual action accounts for its high GH-release potency relative to earlier GHRP compounds. Importantly, somatostatin-mediated negative feedback remains intact, meaning GHRP-2 amplifies natural GH pulses rather than creating sustained supraphysiological GH elevation.

A regulatory note of significance: GHRP-2 (as pralmorelin) has been approved in Japan for clinical use as a diagnostic agent for growth hormone deficiency, providing a regulatory precedent and documented human safety profile. It has not been approved for therapeutic use in the United States, EU, or UK. Its established human data and dual GHS-R/GHRH synergy profile have made it a standard comparator in GH secretagogue research.

Mechanism: GHS-R1a agonist. Activates calcium influx via IP3/DAG pathway, triggering GH granule exocytosis. Also suppresses hypothalamic somatostatin. Evidence suggests GHRP-2 requires functional endogenous GHRH signalling to fully stimulate somatotroph proliferation.

Research Context: Approved in Japan (pralmorelin) for GH deficiency diagnostic testing. Studied in human clinical trials for GH deficiency, paediatric short stature, and appetite/food intake research. Approved comparator in GH secretagogue research programmes worldwide.

Published Research

The following summaries are drawn from peer-reviewed publications indexed on PubMed. GHRP-2 has both extensive preclinical data and human clinical trial data across multiple applications. Pure Grade Labs does not interpret these findings as evidence of efficacy for any unapproved therapeutic indication.

Journal of Clinical Endocrinology & Metabolism: Effects of eight months' treatment with graded doses of GHRP-2 in GH-deficient children

Journal of Clinical Endocrinology & Metabolism: GHRP-2, like ghrelin, increases food intake in healthy men via GHS-R activation

Journal of Neuroendocrinology: GHRP-2 (KP-102) acts through a distinct receptor from GHRH and is 10-fold more potent than earlier GHRP generations in ovine pituitary cells

Certificate of Analysis

Every batch of GHRP-2 supplied by Pure Grade Labs is accompanied by a Certificate of Analysis (COA) from an independent third-party analytical laboratory. The COA confirms identity, purity by HPLC, and absence of contaminants. Batch-specific documentation is linked below.

Storage Requirements

GHRP-2 is supplied as a lyophilised solid. As a hexapeptide (817.97 g/mol), it is reasonably stable in lyophilised form but sensitive to moisture, repeated freeze-thaw cycles, and light. Follow the specifications below to preserve compound integrity throughout the research period.

Temperature: -20°C Long-term storage. For short-term use (up to 4 weeks), 2-8°C is acceptable.

Light Sensitivity: Light-Sensitive. Store in original opaque vial. Avoid direct UV or prolonged exposure to light.

Shelf Life: 24 Months. Lyophilized, sealed, stored at -20°C. Batch expiry printed on vial label.

Preparation Notes

  1. Allow the sealed vial to reach room temperature before opening to minimise moisture introduction.
  2. Use bacteriostatic water (BAC water) as the reconstitution solvent. Add solvent slowly down the inside wall of the vial - do not inject directly onto the lyophilised cake.
  3. Gently swirl the vial until fully dissolved. Do not vortex or shake vigorously. GHRP-2 dissolves readily at room temperature.
  4. Once reconstituted, store at 2-8°C. Use within 28 days. Do not freeze reconstituted solution to avoid peptide degradation from freeze-thaw cycling.
  5. Discard if solution appears cloudy, discoloured, or contains particulate matter.
