Overview

MOTS-c (Mitochondrial Open Reading Frame of the 12S rRNA Type-C) is a 16-amino acid peptide encoded by the mitochondrial genome - specifically within the 12S rRNA gene region of mitochondrial DNA, a location not previously understood to produce bioactive peptides. It was first characterised in 2015 by a research team led by Changhan Lee at the USC Davis School of Gerontology. MOTS-c belongs to a class of compounds known as mitochondrial-derived peptides (MDPs), alongside humanin and SHLP1-6, representing a newly defined category of mitochondrial retrograde signalling molecules.

Unlike nuclear-encoded peptides, MOTS-c is produced within the mitochondria and has been shown to translocate to the nucleus under metabolic or physical stress, where it regulates the expression of stress adaptation-related genes. Its primary signalling pathway involves the Folate-AICAR-AMPK axis - MOTS-c inhibits the folate cycle via methylenetetrahydrofolate dehydrogenase (MTHFD), which triggers AMPK activation as a secondary metabolic response. This mechanism positions MOTS-c as a novel intracellular and endocrine-like metabolic regulator, with documented systemic effects on glucose metabolism, insulin sensitivity, inflammation, and physical performance in preclinical models.

Origin: Encoded within the mitochondrial genome (MT-RNR1 gene). Endogenous MOTS-c levels are elevated by exercise and metabolic stress, and decline measurably with age. Circulating levels in young individuals have been reported to be 11-21% higher than in middle-aged and older populations.

Research Context: Studied primarily in animal models and in vitro systems for its effects on glucose metabolism, insulin resistance, physical performance, aging, and inflammation. No completed human clinical trials have been published as of April 2026. MOTS-c is explicitly banned by WADA beginning in 2024.

Storage Requirements

MOTS-c is supplied as a lyophilised (freeze-dried) solid. As a larger 16-amino acid peptide containing methionine and tryptophan residues, MOTS-c requires careful handling to prevent oxidative degradation. Follow the specifications below for optimal compound integrity.

Temperature: -20°C Long-term storage. For short-term use (up to 4 weeks), 2-8°C is acceptable.

Light Sensitivity: Light-Sensitive. Store in original opaque vial. Avoid direct UV or prolonged exposure to light.

Shelf Life: 24 Months. Lyophilized, sealed, stored at -20°C. Batch expiry printed on vial label.

Preparation Notes

  1. Allow the sealed vial to reach room temperature before opening to minimise moisture introduction from condensation.
  2. Use bacteriostatic water (BAC water) as the reconstitution solvent. Add solvent slowly down the inside wall of the vial - do not inject directly onto the lyophilised cake.
  3. Gently swirl the vial until fully dissolved. MOTS-c is water-soluble and dissolves readily at room temperature. Do not vortex or shake vigorously.
  4. Once reconstituted, store at 2-8°C and protect from light. Use within 28 days. Do not freeze reconstituted solution.
  5. Discard if solution appears cloudy, discoloured, or contains particulate matter.
