Overview

5-Amino-1-Methylquinolinium (5-Amino-1MQ) is a small-molecule research compound developed as a selective inhibitor of nicotinamide N-methyltransferase (NNMT) - an enzyme expressed at high levels in adipose tissue, liver, kidney, brain, and skeletal muscle. When NNMT activity is elevated - a state associated with obesity and metabolic dysregulation - this process consumes NAD+ precursors and methyl donors, impairing mitochondrial function and promoting adipogenic signalling. 5-Amino-1MQ is distinguished by two key pharmacological properties: high membrane permeability (allowing intracellular access) and high selectivity (it does not significantly inhibit related SAM-dependent methyltransferases), making it a useful research probe for isolating NNMT-specific pathway effects.
