Overview

AICAR (5-Aminoimidazole-4-Carboxamide Ribonucleoside), also known as Acadesine, is a naturally occurring nucleoside intermediate in the purine biosynthesis pathway. It is taken up into cells via adenosine transporters and phosphorylated by adenosine kinase to generate ZMP (AICAR monophosphate) - a structural analogue of AMP that directly activates AMP-activated protein kinase (AMPK) by binding to the regulatory gamma subunit at site 3. This mechanism distinguishes AICAR from many AMPK activators that work indirectly by inhibiting mitochondrial function and altering the cellular AMP:ATP ratio.

AMPK functions as a cellular energy sensor. When activated, it switches off anabolic (energy-consuming) processes and switches on catabolic (energy-producing) pathways - inhibiting lipogenesis and fatty acid synthesis while promoting fat oxidation, glucose uptake, and mitochondrial biogenesis. AICAR's ability to pharmacologically recapitulate aspects of this exercise-induced signalling state has made it one of the most extensively used research tools in metabolic biology. It has been studied in clinical settings for cardiac protection, and its endurance-mimicking properties were sufficiently well-established that it was banned by WADA before any formal doping cases were confirmed.

Mechanism: Converted intracellularly to ZMP, which binds the AMPK gamma subunit and activates AMPK. Unlike mitochondrial inhibitors (e.g. metformin), AICAR activates AMPK without directly altering oxygen uptake or the cellular AMP:ATP ratio, allowing more mechanistically specific AMPK research.

Research Context: One of the most widely studied AMPK activators in the literature. Evidence spans rodent models, in vitro systems, and limited human clinical trials (primarily in cardiac contexts). The compound's exercise-mimetic properties are well-documented preclinically but have not been established in controlled human performance trials.

Storage Requirements

AICAR is supplied as a lyophilised (freeze-dried) solid. This form ensures compound stability during transit and long-term storage. Correct handling is required to preserve integrity. Follow the specifications below.

Temperature: -20°C Long-term storage. For short-term use (up to 4 weeks), 2-8°C is acceptable.

Preparation Notes

  1. Allow the sealed vial to reach room temperature before opening to minimise moisture introduction from condensation.
  2. AICAR is freely soluble in water. Reconstitute with sterile water for injection or PBS (phosphate-buffered saline) as appropriate for the experimental system. Add solvent slowly down the inside wall of the vial.
  3. Gently swirl until fully dissolved. Do not vortex or shake vigorously. AICAR dissolves readily and should not require extended mixing.
  4. Once reconstituted, store at 2-8°C. Use within 14 days. Do not freeze reconstituted solution as this may alter stability.
  5. Discard if solution appears discoloured or contains particulate matter.
