Melanotan II (MT-II) is a synthetic cyclic lactam analogue of alpha-melanocyte-stimulating hormone (alpha-MSH), a peptide hormone that occurs naturally in the human body. It was originally developed at the University of Arizona during the 1980s and 1990s, as part of a research programme investigating whether pharmacological tanning could reduce UV-induced skin damage. The compound is a truncated derivative of Melanotan I (afamelanotide), re-engineered into a cyclic structure to enhance stability and receptor activity.

MT-II functions as a non-selective agonist at melanocortin receptors MC1R, MC3R, MC4R, and MC5R. Its cyclic lactam structure confers significantly greater in vivo stability than the linear alpha-MSH precursor, as well as enhanced blood-brain barrier permeability relative to other melanocortin analogues. This multi-receptor profile has made MT-II a widely used molecular tool for investigating melanocortin receptor signalling pathways across pigmentation biology, neuroendocrine research, metabolic regulation, and immune signalling models.

Melanotan II is supplied as a lyophilised (freeze-dried) solid. The cyclic lactam structure of MT-II confers greater stability than many linear peptides, but correct storage conditions remain essential to preserve compound integrity and prevent oxidative degradation of sensitive residues.

Temperature: -20°C Long-term storage. For short-term use (up to 4 weeks), 2-8°C is acceptable.

Light Sensitivity: Light-Sensitive. Store in original opaque vial. Avoid direct UV or prolonged exposure to light.

Allow the sealed vial to reach room temperature before opening to minimise moisture introduction.
Use bacteriostatic water (BAC water) as the reconstitution solvent. Add solvent slowly down the inside wall of the vial - do not inject directly onto the lyophilised cake.
Gently swirl the vial until fully dissolved. Do not vortex or shake vigorously. MT-II dissolves readily in aqueous solution due to its polar cyclic structure.
Once reconstituted, store at 2-8°C and protect from light. Use within 28 days. Do not freeze reconstituted solution.
Discard if solution appears cloudy, discoloured, or contains particulate matter.
